Las fluoropirimidinas son un grupo de agentes citostáticos empleados en el tratamiento de cáncer, como el 5-fluorouracilo (5-FU). El 5-FU es un agente quimioterapéutico usado en tratamientos de distintos tipos de cáncer, como el cáncer de mama, colorrectal, de estómago o pancreático. Es metabolizado por la enzima dihidropirimidina deshidrogenasa (DPD), que es codificada por el gen DPYD. Existen mutaciones de dicho gen que pueden afectar a la actividad de la enzima, acrecentando así el riesgo de toxicidad 5-FU severo o mortal en estos pacientes, de forma que la detección de deficiencias de DPD nos va a ayudar en un pronóstico más preciso de la toxicidad y de su respuesta quimioterapéutica.
Fluoropyrimidines are a group of cytostatic agents used in the treatment of cancer such as 5-fluorouracil (5-FU). 5-fluorouracil (5-FU) is a chemotherapy agent used to treat different types of cancers, including breast, colorectal, stomach and pancreatic cancer. 5-FU is metabolized by the dihydropyrimidine dehydrogenase enzyme (DPD) which is cobed by the DPYD gene. Several mutations within DPYD can affect enzymatic activity, increasing the risk of serious or fatal 5-FU toxicity in these patients, therefore, implementation of DPD deficiency screening by genotyping will allow a more accurate prediction of toxicity and chemotherapeutic response.
